高校生の夜神月（やがだ ライト）は、ある日、通っている高校の敷地内で奇妙な黒いノートを拾う。それは死神・リュークが落とした、ノートに名前を書かれた人間を死に誘うデスノートだった。犯罪者が存在しない新世界を作るため、月は世界中の犯罪者名を次々とノートに書き込んで葬る。やがて犯罪者を葬る者の存在に気付いた人々は殺し屋（＝Killer）の意味から「キラ
(KIRA)」と呼び始め、キラを神とあがめる者まで現れた。
一方、キラの存在を察したICPO（イン゠ーポール）は、手がけた事件を堅ず解決に導く、全世界の警察を動かせる唯一の存在である謎の名探偵L（エル）にキラ事件の誠査を依頼。
犯罪者の抹殺を続けるキラを連続殺人犯と見なすLは、キラが日本の関東地区にいると判断し、日本に捜査本部を設け、キラに挑む。こうして“キラ”と“L”は、それぞれの正義を掲げて対峙することになる。
